SRX21956805: ONT R10.4.1 WGS of Scaptomyza caliginosa: adult female single fly
1 OXFORD_NANOPORE (PromethION) run: 7.1M spots, 7.2G bases, 5.7Gb downloads

Design: Genomic DNA was extracted from adult Drosophila specimens by phenol-chloroform. Oxford Nanopore R10.4.1 libraries were prepared with 1/2 reaction volumes using the LSK114 kit. Basecalling was performed with Guppy 6 with model dna_r10.4.1_e8.2_400bps_sup.cfg with default quality filtering and adapter trimming on.

Study: Genome sequencing and assembly of many species of Family Drosophilidae Genome sequencing and assembly
show Abstracthide Abstract
This work is a collaborative project aiming to broaden the scope of genomic resources for studying evolution in Family Drosophilidae. Sequencing was performed on a variety of sources, ranging from pools of flies from an inbred line to fresh and ethanol-fixed single wild flies. A Nanopore and Illumina hybrid assembly approach was taken to build the genome assemblies. For inbred lines, initial assemblies were generated from Nanopore sequences, polished with Nanopore reads, and finally with paired-end Illumina reads. For individual flies, diploid assemblies were generated from Nanopore reads that were corrected in a haplotype-aware manner with Illumina data.
